Intended Use

Clostridium Perfringens is an anaerobic bacterium commonly found in animal habitats. It metabolizes sugars in organic matter to produce large amounts of gas and toxins, leading to tissue necrosis, toxicosis, and potentially death in animals. In sheep and goats, it can cause lamb dysentery, enterotoxemia and other diseases, severely impacting the sheep farming industry.

This assay is designed to detect antibodies against Clostridium Perfringens (CLP) Alpha-Toxin in the sheep and goats. It can be utilized for the evaluation of the immunological efficacy.

Principle and Application

This kit comprises a coated Microtiter Plate with CLP α-toxin proteins (as the antigens), enzyme conjugates, and other accompanying reagents. It employs the principle of enzyme-linked immunosorbent assay (ELISA) to detect anti-α-toxin antibodies in the ovine/caprine serum or plasma. During the experiment, control serum and test sample are added to the plate. After incubation, if the sample contains anti-α-toxin antibodies, they will bind to the antigens coated on the microtiter plate. Following washing steps to remove unbound components, the enzyme conjugates are added, which specifically binds to the antigen-antibody complexes on the plate. After washing again to remove unbound enzyme conjugates, substrate reagents are added to the wells and react with the enzyme-labeled complexes, resulting in a blue color. The intensity of the color is directly proportional to the amount of specific antibody present in the sample. The reaction is then terminated by adding a stop solution, turning the solution yellow. The absorbance of each well is measured at a wavelength of 450nm using a microtiter plate reader (microplate reader) to determine the presence of antibodies against CLP α-toxin in the sample.
